And if you can't go to Pittsburgh without riding one of the inclines . It's kind of like going to San Fransisco and riding a cable car. I recommend taking the Duquesne Incline - it has free parking at the bottom and offers an amazing view of downtown.
The Natural History Museum is a must, they just renovated Dinosaur Hall, if I'm not mistaken you can get timed tickets for that exhibit in advance and save yourself some hassle - I recommend checking out the Carnegie Museums website to see if you're interested in their other museums.

As far as hotels go I'd say you should stay downtown. The Renaissance is the nicest in the city but I've heard that the Double Tree is the best value. It has a pretty central location and I believe it was just turned into a hotel over the past 2 years so everything is new.

I seem to recall a pretty good art museum. Resturants by the river with trolley's the go up steep hills by the river.
